Identification of S-phase Cells with PCIO Antibody to Proliferating Cell Nuclear Antigen

چکیده

We estimated the expression of proliferating cell nuclear antigen (PCNA) in HeLa S3 cells by flow cytometry with monoclonal antibody (MAb) PC10. HeLa cells were fued with six Merent fuation procedures: 15-mk and 30-min acetone, 15-mb acetone followed by 15-mb methanol (acetonelmethanol), 30-mb methanol, 15-min methanol followed by 15-mh acetone (methanollacetone), and a mixture of acetone and methanol. The fued cells were applied to MAb PClO against PCNA and then treated with FIX. With five fixation procedures except for acetonelmethanol, FCNA was expressed in almost all cells with similar shapes and different IFITC intensity levels on PCNAlDNA bivariate cytograms, whereas acetonelmethanol fixation allowed
